Anatomical aspects of IBA-treated microcuttings of Gomphrena macrocephala St.-Hil BABT

Gomphrena macrocephala St.-Hil. (Amaranthaceae) is a perennial herb from the cerrado with medicinal properties and ornamental interest. Plants can be micropropagated through nodal segments, but acclimatization is difficult. The aim of this study was to establish a relationship between anatomical aspects of the roots and the acclimatization process. When cultures were supplemented with IBA, callus and thick and frangible roots appeared at the base of the microcuttings. A fragile vascular connection between roots and shoots was observed. Abnormal adventitious roots showing alteration in the vascular cylinder and hypertrophy of the cortical cells were also noted. These roots interfere in the transfer to extra vitrum conditions. ABSTRACTThe importance in folk medicine, combined to threats in their environment, becomes necessary to carry out studies involving large-scale propagation of Lippia genus. Although the tissue culture propagation is widely disseminated for medicinal plants, for L. rotundifolia any article was published yet. The present study aimed to establish an efficient protocol for micropropagation of L. rotundifolia. Nodal segments, taken from plants collected in the Espinhaço Range, were disinfected, and cultures were initiated on MS medium with PVPP (1 g L- 1), sucrose (3%) and agar (0.7%). Cedrela odorata L. is one of the most important timber species currently traded in the Caribbean and Central America; however, it has been intensively exploited. In vitro techniques and clonal propagation can help to develop new plantations and assist in establishing improvement programs for this species. The aim of this study was to develop a protocol to establish in vitro conditions and to micropropagate this species from nodal explants from juvenile cuttings taken from field trees. Disinfection of node explants with 5% propiconazole CE 25 during 3 min resulted in 100% explant disinfection and 60% morphogenic response on those established explants.
